Colossatron is a tower offense game where you pick up 'cores' scattered across the screen into your Colossatron in order to equip them. Cores comes in three colors, which can be combined to form secondary colors. When three of the same core are joined together, they combine to become an upgraded core. Cores can be upgraded twice, forming an ultimate core.

The movement and combat of the game is done automatically, which allows players to place cores in their correct positions. This is important as special support cores such as shield and rapid fire is vital to winning stages in the later levels of the game.

Outside of main gameplay, there is a lot for players to do as they have to decide which weapon should be assigned to which color. Each weapon has their own unique range and firepower, and have to be considered carefully as this will impact placement strategy. The game ends after 28 levels, but there is a prestige mode that adds in additional boss fights and harder enemies.
